Effect of water stress on the behavior of cultivars of Coffea arabica under in vitro conditions Efecto del estrés hídrico sobre el comportamiento de cultivares de Coffea arabica bajo condiciones in vitro Ramos-Navas, Ramón Antonio Shimber, Tesfaye Martínez-Suárez, Felipe Taye, Endale Tefera, Wondyifraw Gebre, Endale Mengecha, Ayelign osmotic stress water stress tissue culture embryos cultivation estrés osmótico estrés hídrico cultivo de tejido cultivo de embriones The investigation was developed in the biotechnology laboratory in the Centro de Investigaciones Agrícolas of Gima (Jimma Agricultural Research Centre), to the Southwest of Ethiopia; between January and October of the 2007 period; to value the effect of the osmotic stress (water stress) on the growth of the stalk and roots of four cultivars of Coffea arabica under cultivation conditions in vitro. Samples of mature seeds of four Coffea arabica cultivars were used (Aba Buna, F-59, 7487 and Gawe), taken place in the season from the 2006 to the 2007 and benefitted by Coffee Breeding department and Coffee Agronomy of JARC. The zygotic embryos were deposited on the basal means SH (Schenk and Hildebrandt, 1972), containing BAP (0.02 mg/L), ELL (0.02 mg/l) and GA3 (0.05 mg/L). Six concentrations of the Manitol osmotic agent’s (0, 2, 4, 6, 8 and 10 %) were used for each coffee genotype. A significant differences among the coffee germplasm subjected to the stress (p < 0.05) for the root longitude. The growth of the root of the four cultivars was significantly inhibited with the osmotic stress taken place by the use of the Manitol. However, a Manitol concentration to 2 % produced the stimulation in the elongation from the root to the 20 days and 45 days after the treatment. The lethal doses (DL) for the root growth were 6 % from Manitol to the 20 days and 6.5 % from Manitol to the 10 days and 45 days.  La investigación se desarrolló en el Laboratorio de Biotecnología en el Centro de Investigaciones Agrícolas de Gima (Jimma Agricultural Research Centre), al suroeste de Etiopia; en el período comprendido entre enero y octubre de 2007, con el objetivo de evaluar el efecto del estrés osmótico (estrés hídrico) sobre el crecimiento del tallo y raíces de cuatro cultivares de Coffea arabica bajo condiciones de cultivo in vitro. Se utilizaron muestras de semillas maduras de cuatro cultivares de Coffea arabica (Aba Buna, F-59, 7487 y Gawe) producidas en la temporada de 2006 a 2007 y beneficiadas por el departamento de Coffee Breeding y Coffee Agronomy de JARC. Los embriones cigóticos se depositaron sobre el medio basal SH (Schenk and Hildebrandt, 1972), conteniendo BAP (0,02 mg/L), ANA (0,02 mg/L) y GA3 (0,05 mg/L). Se utilizaron seis concentraciones del agente osmótico Manitol (0, 2, 4, 6, 8 y 10 %) por cada genotipo de café. Se encontraron diferencias significativas entre los germoplasmas de café sometidos al estrés (p < 0,05) para la longitud de la raíz. El crecimiento de la raíz de los cuatro cultivares fue significativamente inhibido con el estrés osmótico producido por el uso del Manitol. Sin embargo, una concentración de Manitol al 2 % produjo la estimulación en la elongación de la raíz a los 20 días y 45 días luego del tratamiento. Las dosis letales (DL) para el crecimiento de la raíz fueron del 6 % de Manitol a los 20 días, y 6,5 % de Manitol a los 10 y 45 días.
